dc.description.abstractLos oligodeoxinucleótidos Citocina- Guanina (ODN CpG) son secuencias sintéticas cortas de ADN que contiene dinucleótidos CpG no metilados reconocidos a través del Receptor Tipo Toll-9 (TLR9), que han demostrado activar el sistema inmune de peces al ser administrados vía intraperitoneal, sin presentar daño histopatológico asociado a la inmunoestimulación. Sin embargo este efecto depende de la longitud de la secuencia, el número de motivos CpG, posición, espaciado y las bases circundantes de estos motivos, catalogándolos como especie-especifico. Actualmente, el estudio de los órganos asociados a la producción de mucus en peces (como la piel, branquias, nasofaringe e intestino) ha ido en incremento, por ser el primer contacto entre agentes patógenos y huésped, resultando ser una de las primeras barreras del sistema inmune. Sin embargo, se desconoce si los ODN CpG pueden tener un efecto inmunoestimulante en este tipo de tejido en peces, específicamente en la producción de mucus. Por tanto, el objetivo de esta investigación fué evaluar el efecto inmunoestimulante de diferentes ODN CpG sintéticos en el pargo amarillo (Lutjanus argentiventris). Se llevó a cabo un experimento in vivo con juveniles (L. argentiventris), donde se evaluaron 7 diferentes tratamientos (inyección intraperitoneal), cinco tratamientos con diferentes CpGs, un tratamiento con Vibrio parahaemolyticus + PBS y un control con PBS. Se realizaron cuatro muestreos (día 1, 3, 7 y 10) de hígado, bazo e intestino. Se determinó la actividad enzimática antioxidante, expresión de genes inmunorelevantes, el análisis histológico de intestino por microscopia óptica, así como la cuantificación de mucinas ácidas y neutras. Los resultados obtenidos demostraron que los ODN CpG1826, 1670 y 2006, tienen la capacidad de modular la expresión tanto en bazo como intestino de interlucinas proinflamatorias IL-β e IL-8 (4 veces más que el control), factores de transcripción asociados a la maduración de células T y B (ILF2/NF45) y producción de interferón (IRF7). Así mismo, se observó un incremento en la producción de mucinas neutras sin registro de daños histopatológicos en intestino; no se observó un efecto significativo en la actividad de enzimas antioxidantes (SOD, CAT) descartando un posible efecto por estrés. Así mismo, pudimos comprobar que uno de los principales componentes que afecta la respuesta inmune de L. argentiventris por la administración vía intraperitoneal de ODN CpG, tanto en bazo como en intestino, son los factores humorales asociados a la respuesta pro-inflamatoria. Sin embargo, no se observó la sobrexpresión de TLR9 y NFκβ, sugiriendo un posible efecto de regulación negativa en la expresión o el reconocimiento por el TLR21, un homólogo del patrón de reconocimiento de TLR9 en peces y aves.es_MX

dc.description.abstractenCytosine-Guanine oligooxynucleotides (CpG ODN) are synthetic short-DNA sequences with non-methyled CpG motifs recognized by Toll-like receptor 9, has improved the response of immune system when are intraperitoneally administered, without histopathological injury associated to immunostimulation. However, their actions are depending of CpG motifs spacing, position, surrounding bases and sequence length, being species-specific. Recently research of organs associated production mucus has been increasing, for being the first interaction between pathogens and host considering it an immune barrier. Nerveless, the imunoestimulatory effect by the CpG ODN on this type of fish organs are unknown, specilly mucus production. Therefore, the objective of this research was to evaluate the imunostimulatory effect of different synthetic ODN CpG on yellow snapper (Lutjanus argentiventris). Five CpG ODN and Vibrio parahemolitycus were administrated by intraperitoneally injection on juveniles yellow snapper, PBS was use as a negative control. Four sampling (day 1, 3, 7, 10) of liver, spleen and gut were performed. Antioxidant enzymatic activity, quantification of immunorelevant genes, intestinal histological analysis by optical microscopy, as well as the quantification of acid and neutral mucins were determined. The results obtained demonstrated that CpG ODNs 1826, 1670 and 2006 have the ability to modulate spleen and gut expression of proinflammatory IL-β and IL-8 interleukins (4 times more than control), and associated transcription factors on T and B cells maturation (ILF2 / NF45) and interferon production (IRF7). A neutral mucin increase was also observed, without histopathological intestine damage. There was no significant effect on antioxidant enzymes activity (SOD, CAT) ruling out a possible stress effect. Likewise, we verified through a factorial analysis that one of the main components that affects the immune response of L. argentiventris by the intraperitoneal administration of CpG ODN in spleen and intestine are the humoral factors associated with the pro- Inflammatory response. No overexpression of TLR9 and NFκβ was register, suggesting a TLR9 expression negative regulatory effect or recognition by TLR 21, a homologue recognition receptor TLR9 on fish and birds.es_MX
