dc.description.abstract"La enfermedad hepatopancreática aguda (AHPND) es causada por la bacteria Vibrio parahaemolyticus y es un severo problema en granjas de camarón a nivel mundial. El material infectivo de V. parahaemolyticus que causa la AHPND son las toxinas PirABvp codificados en un plásmido. El tejido diana de la enfermedad es el hepatopáncreas, aunque el estómago también tiene afectaciones. Los signos de la AHPND son: el desprendimiento de células epiteliales, la necrosis, la infiltración de hemocitos y la atrofia celular. En esta tesis se aborda principalmente que la AHPND causa afectaciones en el metabolismo y sistema inmune del camarón. Por lo que la sección experimental abordó 4 secciones: I) evaluación y caracterización de la cepa V. parahaemolyticus IPNGS16 y su concentración letal 50, II) Evaluación de lesiones y el progreso de la AHPND mediante histología, III) Evaluación de variables bioquímicas en hepatopáncreas, músculo y hemolinfa durante la AHPND, IV) Evaluación de la expresión global y la expresión relativa de genes en hepatopáncreas y en hemolinfa mediante ARNm-Seq y RT-qPCR. La cepa de V. parahaemolyticus IPNGS16 contiene el plásmido que codifica a las toxinas PirABvp, y se determinó que a menor dosis menor mortalidad, a mayor dosis mayor mortalidad y se obtuvo la LC50 a las 24 horas post-infección (hpi). Las lesiones evaluadas por histología muestran que los signos de la AHPND aparecen a las 12 hpi y se agravan a las 24 hpi de manera significativa. Las variables bioquímicas en los tres tejidos fueron diferentes y dinámicas de acuerdo con el paso del tiempo. Los reportes de la patogénesis de la AHPND nos permiten relacionar 2 eventos, el primero con la infección de V. parahaemolyticus y el segundo con el efecto de las toxinas. Finalmente, la expresión global de genes en hepatopáncreas nos permitió entender y ligar procesos biológicos enriquecidos relacionados con la respuesta inmune, metabólica, y adhesión biológica. Mientras que con la expresión global en hemocitos logramos identificar el modelo de respuesta a insecticidas ejemplificando el dicloro difenil tricloroetano (DDT). La expresión relativa de genes del sistema antioxidante, hemocianina, coagulación entre otros, nos ayudó a corroborar la relación con los datos de ARNm-seq. La investigación y los datos obtenidos nos otorgan las pruebas para demostrar que existe un cambio metabólico e inmune en L. vannamei durante la AHPND a nivel expresión de genes y los cambios de las variables bioquímicas..."es

dc.description.abstracten"Acute hepatopancreatic necrosis disease (AHPND), caused by the marine bacteria Vibrio parahaemolyticus, is a considerable problem in shrimps farms. The V. parahaemolyticus infecting material is contained in a plasmid that encodes for the lethal toxins PirABvp, whose primary target tissue is the hepatopancreas, causing sloughing epithelial cells, necrosis, and massive hemocyte infiltration. This work is focused on the changes in the metabolic and immune systems caused by AHPND. The experimental section is divided into four sections: I) To evaluate and characterize the V. parahaemolyticus IPNGS16 strain and the lethal concentration 50. II) To evaluate lesion and progression of AHPND using histological techniques. III) To assess biochemical parameters in hepatopancreas, muscle, and hemolymph during AHPND. IV) To assess the global gene expression in hepatopancreas and hemocytes at 24 hours post-infection (hpi) using ARNm-Seq and RT-qPCR. The V. parahaemolyticus IPNGS16 strain contains the plasmid that encodes the PirABvp toxins; using different bacteria concentrations, this strain shows dose-response toxicity, the higher concentration of bacteria, the higher the mortality in less time, and a low bacteria concentration results in less mortality. Histological evaluation of infected hepatopancreas indicates that the main signs of AHPND appear at 12 hpi and significantly worsen by 24 hpi. The biochemistry parameters acquired from the three tissues tested showed a dynamic and tissue-specific AHPND progression through time, although these results suggest relating to events, the first relate-bacterial infection and the second with the toxin-neutralization. Furthermore, the global gene expression analysis in the hepatopancreas reveals crosstalk between three enriched biological processes, the immune, metabolic, and biological adhesion processes. On the other hand, the global gene expression observed in hemocytes reflects a response like the insecticides model, more specifically to the DDT response. These findings deepen our understanding of the hepatopancreas and hemocyte response and the relative gene expression (mRNA levels) results obtained, validate, and corroborate the RNAseq data. This study shows that in the AHPND, the bacterial infection and the effect of toxins should be seen as the same event rather than two independent processes. Indeed, the results presented here increase our understanding of the immune- metabolic response of L. vannamei towards AHPND."es
