dc.description.abstract"La colecta de muestras biológicas en campo puede ser difícil. El uso del papel filtro de Nobuto es una alternativa ya que permite la colecta de fluidos biológicos de manera simple y rápida. Se colectaron muestras de sangre y leche de bovinos (n=11) y caprinos (n=27) provenientes de 3 ranchos de Todos Santos, Baja California Sur, México. La sangre se obtuvo por extracción venosa yugular o coccígea en tubos Vacutainer® con heparina como anticoagulante y la leche se obtuvo por ordeña manual en tubos de plástico estériles. Tiras de papel filtro Nobuto® se saturaron con sangre o leche y se dejaron secar al aire. Previo al análisis, cada tira se sumergió en solución salina y se eluyó durante la noche en refrigeración. Se usaron los datos de las muestras de sangre y leche colectadas por métodos tradicionales como estándares. Todas las muestras se procesaron para la detección de anticuerpos contra Brucella abortus y Anaplasma spp. por medio de pruebas serológicas, y para la cuantificación de la concentración de proteínas totales, glutatión (GSH-Eq, GSH, GSSG), carbonilos proteicos y sustancias reactivas al ácido tiobarbitúrico (TBARS) como indicadores de estrés oxidativo. No se detectaron anticuerpos para Brucella abortus y Anaplasma spp. en las muestras de leche y sangre; por lo tanto, no se analizaron las muestras del papel filtro. Los resultados sugieren que los animales incluidos en este estudio están sanos y libres de brucelosis y anaplasmosis. Se detectaron indicadores de estrés oxidativo por primera vez en papel filtro con una frecuencia relativa >75%. Las concentraciones de GSH-Eq, GSH, GSSG, TBARS y carbonilos proteicos preservados en el papel filtro de Nobuto no fueron comparables con su estándar de referencia. Las muestras de sangre preservadas en el papel filtro se pueden usar para la evaluación del estado redox por medio de la tasa GSSG:GSH-Eq. El papel filtro de Nobuto es un método apropiado para analizar moléculas más estables."es

dc.description.abstracten"Collecting biological samples at remote field sites with limited processing capabilities can be problematic. The use of Nobuto filter paper is an alternative to collect samples in different environmental conditions, contributing to facilitate clinical, research and biomonitoring efforts. Drying the filter paper facilitates transport and allows for simple storage. The objective of this study was to validate the use of Nobuto filter paper for the collection of blood and milk from livestock. Blood and milk samples were collected from cattle (n=11) and goats (n=27) in 3 ranches of Todos Santos, Baja California Sur, Mexico. Blood was obtained by jugular or coccygeal venipuncture with heparin as an anticoagulant. The milk was obtained by manual milking in sterile plastic tubes. From each sample, an aliquot was collected in Nobuto filter paper by soaking 15 strips per sample. The filter paper was air dried at room temperature and stored in labeled envelopes for shipping and further analysis. The remaining samples were processed following conventional methods and frozen for transport and subsequent analysis. Prior to analysis, each filter paper strip was immersed in saline and eluted overnight under refrigeration. All samples were processed following the same methods for the detection of antibodies against Brucella abortus and Anaplasma spp. by serological tests, and quantification of the concentrations of total proteins, glutathione (GSH-Eq, GSH, GSSG), protein carbonyls and thiobarbituric acid reactive substances (TBARS) as indicators of oxidative stress. No antibodies were detected for Brucella abortus and Anaplasma spp. in milk and blood samples; therefore, the filter paper samples were not analyzed. The results suggest that the animals included in this study are healthy and free from brucellosis and anaplasmosis. Indicators of oxidative stress were detected for the first time on filter paper with a relative frequency >75%. The concentrations of GSH-Eq, GSH, GSSG, TBARS and protein carbonyls preserved on Nobuto filter paper were not comparable with their gold standard. Blood samples preserved in Nobuto filter paper may be used for the evaluation of the redox status from the GSSG:GSH-Eq ratio. Nobuto filter paper is an suitable method for analyzing more stable molecules."es
