dc.description"La transferencia genética horizontal, también llamada transferencia genética lateral, es un mecanismo importante por medio del cuál las bacterias transmiten e incorporan material genético sin implicar la recombinación genética sexual y que les proporcionan una mayor variabilidad genética como base esencial para la evolución de estas. En la naturaleza los microorganismos frecuentemente intercambian información genética mediante la toma de ADN libre en el medio ambiente (transformación), la introducción de ADN mediada por bacteriófagos (transducción) o transferencia directa de ADN mediante el contacto directo de dos bacterias (conjugación). La conjugación es probablemente el mecanismo más eficiente para transferir información genética, o el menos difícil de estudiar. En este trabajo se estudió la transferencia genética horizontal in vitro vía experimentos de conjugación en el laboratorio e in vivo en un cultivo de trigo inoculado con Azospirillum halopraeferens Au4. No se logró detectar la transferencia genética horizontal de plásmidos por conjugación entre A. halopraeferens Au4 y las poblaciones bacterianas de la rizósfera de trigo in vivo, debido a la dificultad de monitorear la transferencia en ambientes abiertos, a pesar de haber detectado plásmidos pertenecientes al grupo Inc P-1 presentes en casi la totalidad de los tratamientos, los cuales facilitarían el monitoreo de la transferencia in vivo. Sin embargo, se logró detectar in vivo mediante técnicas moleculares la presencia de plásmidos altamente distribuidos, pertenecientes a los grupos de incompatibilidad Inc P-1a e Inc P-1b en la rizósfera de trigo. Por otro lado, en los experimentos de aislamiento exógeno por conjugación triparental, se logró detectar la presencia de plásmidos en la mayoría de los tratamientos, los cuales fueron co-movilizados utilizando el plásmidos pSM1890, obteniéndose frecuencias de conjugación del orden de 10-6-10-7, lo que concuerda con otros reportes usando plásmidos del mismo grupo (Inc Q) al que pertenece el plásmido movilizador. No fue posible realizar conjugación biparental usando A. halopraeferens o A. brasilense como donador, debido a que no teníamos información suficiente acerca de la codificación de marcadores moleculares contenida en los plásmidos de las cepas donadoras."
dc.descriptionHorizontal gene transfer (lateral gene transfer) is the movement of genetic material between bacteria; which is different from gene flow transmitted to the descendants. This important mechanism in bacteria provides greater genetic variability and promotes evolution of these microorganisms. In nature, microorganisms frequently interchange genetic information through free DNA from surrounding microorganisms in the environment (transformation), DNA introduced by bacteriophage infection (transduction), or direct DNA transference by direct mating of two bacteria (conjugation). Conjugation is probably the most efficient way to transfer genetic information, or is possibly easier to study. In this project, horizontal genetic transference in vitro was studied in experiments of conjugation in laboratory and in vivo in wheat culture inoculated with Azospirillum halopraeferens Au4. Horizontal genetic transfer through plasmids was not detected by mating between A. halopraeferens Au4 and bacteria populations of wheat rhizosphere in vivo due to monitoring difficulty of transfer in open environments in spite of having detected plasmids belonging to the group Inc P-1 present in almost the totality of the treatments, which would facilitate monitoring the transfer in vivo. However, Mobile plasmids were detected in vivo using molecular techniques pertaining to the groups Inc P-1a and Inc P-1b in wheat rhizosphere. On the other hand, in the experiments of exogenous isolation by triparental mating, plasmids were detected in most of the treatments, which were co-mobilized using plasmid “pSM1890” with transfer frequencies on the order of 10-6 to 10-7. This agrees with other reports using plasmids of the Inc Q group as mobilizing plasmids. Biparental mating was not developed using A. halopraeferens Au4 o A. brasilense Cd as donor, because of the lack of sufficient information about the codification of molecular markers contained in plasmids of donor strains. However, in triparental mating, using a helper strain which contained a plasmid “pSM1890” a mobile plasmid was detected in A. brasilense Cd as in A. halopraeferens Au4 with an average transfer frequency of 1.15 x10-1. In the case of Azospirillum halopraeferens Au4 as donor strain, the single mobile plasmid was observed only in two of the ten isolated transconjugants, obtaining an average transfer frequency of 7.06 x10-1.
