dc.descriptionLos ácidos grasos poliinsaturados (PUFAs, por sus siglas en inglés), tales como el ácido eicosapentaenoico 20:5 -3 (EPA), se han reconocido por sus usos potenciales en la prevención y el tratamiento de diversas enfermedades. Las algas marinas, tales como la diatomea Phaeodactylum tricornutum son la principal fuente de estos compuestos. El empleo de las mutaciones como sistema de mejoramiento genético tiene un gran potencial para incrementar la producción de ácidos grasos y otros metabolitos en algas. El objetivo del presente trabajo fue la obtención de una cepa mutante de P. tricornutum, sobreproductora de EPA, por mutagenesis química, y su caracterización. Mediante exposición de un cultivo de P. tricornutum al mutágeno etil metano sulfonato (EMS), se generaron cepas mutantes de P. tricornutum con una frecuencia de mutación que osciló entre 13 y 29%. De forma paralela se realizaron pruebas con la cepa silvestre, para establecer parámetros físicos de selección de cepas mutantes con posibles modificaciones en su contenido de ácidos grasos. Se establecieron cambios de temperaturas e intensidad luminosa de incubación como factores de selección de cepas mutantes. El intervalo de temperatura propuesto para este fin fue 4 a 14°C con una intensidad luminosa de 87mol quanta m-2s-1, ya que bajo estas condiciones disminuye la tasa de crecimiento de la cepa silvestre sin que se afecte el contenido de EPA. Como resultado de la mutagénesis, se aislaron aleatoriamente 66 colonias, las cuales fueron crecidas bajo las condiciones de estrés ambiental elegidas previamente. En esta etapa se seleccionaron 19 colonias que presentaron resistencia a los cambios en los parámetros de incubación e incrementaron su tasa máxima de crecimiento con respecto a la cepa silvestre. Cinco meses después de la primera selección sólo 8 cepas conservaron su resistencia al estrés ambiental, mostrando tasas de crecimiento 4 a 9 veces más altas que la cepa silvestre. Las 8 cepas seleccionadas no mostraron diferencias significativas al ser caracterizadas bajo condiciones estándares (20°C y 60mol quanta m-2s-1), en las tasas máximas de crecimiento y la biomasa total respecto a la cepa silvestre. dc.descriptionThe polyunsaturated fatty acids (PUFAs), as the eicosapentaenoic acid 20:5 -3 (EPA), have been recognized as potential therapeutic agents in the treatment and prevention of several ailments. The main source of PUFAs are marine algae, among these the Phaeodactylum tricornutum is one of the most productive PUFAs generator. Induced mutagenesis has been proposed to be used in algae in an attempt to generate stable genetic changes that increase the production of fatty acids and other metabolites. The principal scope of this work was to generate –by chemical mutagenesis—an EPA over producer clone of P. tricorntum, employing ethyl-methane-sulphonate (EMS). A P. tricornutum culture was exposed to different EMS concentrations. At the same time another series of experiments were performed with the P. tricornutum wild type to establish physical parameters as selection factors for mutant clones. In these experiments incubation temperature was dropped from 20˚C to a 4-14˚C range and light intensity was increased from 60mol quanta m-2 s-1 to 87mol quanta m-2 s-1 These conditions were called “environmental stress” (ES). Under these conditions the wild type growth rate was affected without effects on the EPA content. After treatment of P. tricornutum with EMS, clones with a 13–29% mutation rate were obtained. 66 randomly selected EMS induced (possible) mutants were grown under ES conditions. From this group only 19 potentially mutant clones were selected because of their resistance to the ES conditions compared to the wild type. After a 5 month period only 8 mutant candidates were established and their growth rate, under ES conditions, was 4 to 9 fold higher than the wild type. When these 8 clones and the wild type were grown under normal temperature and luminosity conditions, no statistical difference was observed in their growth rate nor in their total biomass. However significant differences were found in their fatty acid content, including EPA and its precursors, when comparing these parameters to the ones obtained from the wild type, suggesting that most likely, permanent changes at the molecular level may have occurred. Moreover, the most interesting change was the EPA and 16:3 increase in the putative mutant clones, which suggested us, that is one of the cell mechanisms that allowed their survival-under-ES-conditions.
