dc.descriptionEn ostiones, la maduración de la gónada se encuentra regulada por cambios de calidad y cantidad de alimento, temperatura, salinidad, y cambios químicos en el agua que incluyen la presencia de esperma, factores que a su vez modulan la síntesis y secreción de hormonas que controlan la reproducción, entre ellas, las prostaglandinas (PGs). Las PGs son derivados de ácidos grasos de 20 carbonos, siendo la más común la PGE2. Las distintas PGs pueden presentar funciones variadas dependiendo de su concentración, el estado fisiológico del organismo y la especie. En invertebrados, se ha descrito que las PGs modulan el sistema inmune y la reproducción, tanto a nivel de gametogénesis como del desove. Uno de los objetivos de la presente tesis fue cuantificar la PGE2 durante el desove y a consecuencia del estrés de muestreo en ostiones. Para cuantificar las PGs, se compararon tres técnicas; cromatografía de líquidos (HPLC-MS) y cromatografía de gases (CG-MS), ambas con detección por espectro de masas, y kit de inmunoensayo (EIA) específico para PGE2. Aún cuando el EIA presentó una ligera reacción cruzada con otras PGs y con el ácido araquidónico, sus ventajas son que no se requiere de tantos cuidados para almacenar y procesar las muestras, su extracto derivatizado es estable, se requiere poca cantidad de muestra y es el método de menor costo. Para establecer el efecto del estrés de muestreo se utilizaron ostiones de Crassostrea corteziensis y C. gigas en diferentes muestreos. El estrés consistió en cortar el músculo aductor para abrir las valvas y muestrear a los organismos a los 0, 3, 5, 10, 30 y 60 min. No se encontraron diferencias significativas a lo largo del tiempo después del estrés de muestreo, sin embargo, la variación individual en los niveles de PGE2 aumentaron a partir del minuto 5, disminuyendo hasta los 60 minutos, por lo que, para futuros muestreos para el análisis de PGs, se recomienda muestrear a cada organismo en un lapso no mayor a 3 minutos después de abiertas las valvas. Los niveles de PGE2 en relación al desove se analizaron en ostiones C. gigas inducidos al desove por medio de incrementos de temperatura (de 18 a 30ºC) y por adición de peróxido de hidrógeno (H2O2, 1 ml/l) en el agua. Los ostiones se muestrearon antes de la inducción, durante el desove y 18-24 horas después del desove. No se encontraron diferencias significativas a lo largo de las fases del desove inducido por incremento de temperatura, sin embargo se observa que los niveles de PGE2.
dc.descriptionIn oysters, gonadic development is regulated by quality and quantity of food, temperature, salinity, and chemical changes in water including sperm presence, these factors modulate synthesis and secretion of hormones involved in reproduction control, such as prostaglandins (PGs). PGs are synthesized from 20 carbon fatty acid, the most common being PGE2. PGs can have different functions in relation to their concentration, the physiological state of the organism and the species. In invertebrates, PGs are known to modulate the immune system and reproduction, including gametogenesis and spawning. One of the aims of this thesis was to quantify the PGE2 during spawning and as a consequence of the sampling stress in oysters. To quantify PGs, three methods were compared; liquid (HPLC-MS) and gas chromatography (CGMS), both with mass spectrometry detection, and immunoassay kit (EIA) specific for PGE2. In spite of some cross-reaction with other PGs and with arachidonic acid using EIA, this method had the advantage that it did not require special management and storage of samples, the derivatized extract is stable, only a small quantity of sample is needed and it is comparatively, less expensive than the other two methods. To establish the effect of sampling stress Crassostrea corteziensis and C. gigas oysters were used in different samplings. The stress consisted in cutting the adductor muscle to open the valves and sampling the oyster at 0, 3, 5, 10, 30, and 60 minutes. No significant differences were found for time after sampling stress, but the individual variation on PGE2 levels increased from 5 min and did not decrease till 60 min. It is recommended that sampling for PG analysis is done in less than 3 min after the valves are opened. PGE2 levels were analyzed in C. gigas induced to spawn with thermal shock (from 18 to 30ºC) and by addition of hydrogen peroxide (H2O2, 1 ml/l) to the water. Oysters were sampled before the induction, during spawning and 18-24 hours after they had spawned. No significant differences were found during the spawning process induced by thermal shock, nevertheless, levels of PGE2 tended to decrease during spawning and even more after 18-24 hours. The induction with H2O2 produced significant difference in PGE2 during spawning in males, but not in females; in both cases the levels of PGE2 increased during spawning and increased further after spawning [...]
