dc.descriptionLa lectina del suero del camarón blanco (L. vannamei) se purificó por cromatografía de afinidad, utilizando estroma de eritrocitos de rata fijados con glutaraldehido. En el análisis de la lectina en gel de poliacrilamida en condiciones desnaturalizantes se identificaron cuatro fracciones de 83.14, 64.01, 46.16 y 33.29 kDa. La lectina aglutina eritrocitos de diferentes especies (conejo, rata, humano, cobayo y hámster) y tiene una mayor actividad aglutinante con los eritrocitos de ratón. La actividad aglutinante aumentó en presencia de Ca++, así como en presencia de eritrocitos tratados con tripsina o con sialidasa. La actividad hemaglutinante fue inhibida por glucosamina, fucosa, glucosa y galactosa. Se evaluó el efecto de dos mezclas de bacterias del género Bacillus (2x106 cel ml-1), de la levadura Debaryomyces hansenii (1x106 cel ml-1) y del inmunoestimulante comercial laminarina (0.5 mg L-1) en la concentración de lectina en la hemolinfa del camarón blanco L. vannamei. Los tratamientos se agregaron directamente al agua de los camarones cada 3 días. El bioensayo tuvo una duración de 14 d, se extrajo hemolinfa con anticoagulante de tres camarones por cada tratamiento a las 24, 48 y 72 h después de aplicados los tratamientos. La concentración de lectina de los camarones se determinó por ELISA, utilizando anticuerpos monoclonales contra la lectina de Macrobrachium rosenbergii (crustácea decápoda). Nuestros resultados confirman que los anticuerpos reconocen epítopes comunes con camarón, pero no se identificaron diferencias significativas entre la concentración de lectina en el plasma de L. vannamei expuestos a diferentes tratamientos y los organismos control sin tratamiento.
dc.descriptionThe lectin from the serum of the shrimp Litopenaeus vannamei was purified by affinity choromatography on glutarldehyde-fixed stroma from rat erythrocytes. Analysis of the lectin by polyacrylamide gel electrophoresis under reductive conditions showed four fractions of 83.14, 64.01, 46.16 and 33.29 kDa. The lectin agglutinates erythrocytes of different species (rabbit, rat, human, guinea pig and hamster) and has the higher agglutinating activity for mouse erythrocytes. The agglutinating activity increases in the presence of Ca++, as well as with erythrocytes treated with tripsin or sialidase. The hemagglutinating activity was inhibited by glucosamine, fucose, glucose and galactose. It was evaluated the effect of two mixes of Bacillus bacteria genus (2x106 cel ml-1), the yeast Debaryomyces hansenii (1x106 cel ml-1) and the commercial immunostimulant laminarin (0.5 mg L-1) on the concentration of lectin in the shrimps´ hemolymph. The treatments were directly added to the shrimps´ water every 3 days. The experiment lasted 14 d; hemolymph was extracted with anticoagulant from three shrimps of each treatment at 24, 48, and 72 h after the treatments. The shrimps´ lectin concentration was determined by ELISA using monoclonal antibodies generated against Macrobrachium rosembergii lectin. Our results confirm that the antibodies recognize common epitopes with shrimp, but we didn’t indentify significant differences between the lectin concentration in the plasma of the treated white shrimps (L. vannamei) and the lectin concentration in the plasma of the control organisms without treatment.
