dc.descriptionSe llevó a cabo el aislamiento, caracterización y evaluación in-vitro e in-vivo de bacterias obtenidas del tracto digestivo de la almeja Pata de Mula Anadara tuberculosa, a fin de determinar su potencial probiótico. Se evaluaron in-vitro 151 cepas, determinando actividad hemolítica, hidrofobicidad, tolerancia a nitrógeno amoniacal, salinidad y pH, cinética de crecimiento, actividad enzimática extracelular, autoagregación y coagregación. Se seleccionaron siete cepas que se identificaron por métodos moleculares, de las que, en Litopenaeus vannamei se evaluaron las cepas MAt29, MAt35, GAtB1, GAtBAL7 y la mezcla de las cuatro. Se registró una diferencia significativa con GAtB1 en el conteo de hemocitos, en relación al grupo control. La mejor tasa de crecimiento específico (TCE) se obtuvo con la mezcla. En un segundo bioensayo se evaluaron diferentes concentraciones de otra mezcla bacteriana (GAtB1, MAt32 y MAt43). La mayor TCE y menor prevalencia de WSSV se obtuvo con la concentración de 106 UFC∙gr-1 de mezcla bacteriana. Los menores porcentajes de prevalencia de IHHNV y WSSV+IHHNV, se obtuvieron a una concentración de 4 ×106 UFC∙gr-1. Se registraron diferencias significativas en la expresión de los genes proFO, SOD, LvToll y HSP70 en L. vannamei con la mezcla bacteriana. Las cepas GAtB1, MAt32 y MAt43 se evaluaron en cultivos de microalgas utilizadas como alimento de bivalvos, no encontrando efecto significativo en el crecimiento de las microalgas. Las cepas GAtB1, MAt32 y MAt42 se evaluaron en ostión Kumamoto Crassostrea sikamea. Se registró un mayor crecimiento en los grupos adicionados con bacterias y menor mortalidad ante un reto con Vibrio parahaemolyticus. Se comprobó la adhesión de dichas cepas al epitelio de C. sikamea, mediante microscopía de fluorescencia. Las bacterias aisladas de A. tuberculosa tuvieron un efecto benéfico sobre el crecimiento y respuesta inmune de L. vannamei y C. sikamea, lo que sugiere que tienen potencial probiótico para su aplicación en la acuicultura de organismos marinos
dc.descriptionIsolation, characterization, and evaluation in-vitro and in-vivo of bacterial strains obtained from the gut of Pata de Mula Anadara tuberculosa were carried out to determine their probiotic potential. A total of 151 strains was evaluated in vitro for hemolytic activity, hydrophobicity, ammonia nitrogen tolerance, salinity and pH, growth kinetics, extracellular enzymatic activity, self-aggregation and co-aggregation, and identified by molecular methods. Strains MAt29, MAt35, GAtB1, GAtBAL7 and their mix were evaluated in Litopenaeus vannamei. A significant difference was recorded in counting hemocytes with GAtB1 compared to the control group. The best specific growth rate (SGR) was obtained with the mixture. In a second bioassay, different concentrations of bacterial mixture (GAtB1, MAt32 and MAt43) were evaluated. The best SGR and lowest prevalence of WSSV were obtained by using a concentration of 106 CFU∙g-1. The lowest percentages of prevalence of IHHNV and WSSV + IHHNV were obtained at a concentration of 4 × 106 CFU∙g-1. The bacterial mixture showed significant differences in the expression of proPO, SOD, LvToll, and HSP70 genes in L. vannamei. Strains GAtB1, MAt32 and MAt43 were evaluated in cultures of microalgae used as food for bivalves with no significant effect found on algal growth. Strains GAtB1, MAt42 and MAt32 were evaluated in Kumamoto oyster Crassostrea sikamea finding better growth in the groups with added bacteria and lower mortality during a challenge against Vibrio parahaemolyticus. Adhesion of the same strains to C. sikamea epithelium was observed using fluorescence microscopy. In general, bacterial isolates of A. tuberculosa showed a beneficial effect on growth and immune response of L. vannamei and C. sikamea, suggesting they have potential as probiotics for application in marine organism aquaculture
