dc.descriptionTomato chino La Paz virus (ToChLPV) y el virus del mosaico dorado del chile (PepGMV, Pepper golden mosaic virus) son begomovirus que se han adaptado a un amplio rango de hospederos y se han encontrado coinfectando, con otros begomovirus, al mismo hospedero, causando enfermedades en los principales cultivos de interés agronómico. El mecanismo de silenciamiento génico o RNA de interferencia (RNAi) se ha utilizado con éxito en el control de enfermedades ocasionadas por begomovirus; las estrategias de ingeniería genética para resistencia a este tipo de virus han contemplado el uso de construcciones con secuencias homólogas al virus retado. Sin embargo, la presencia de infecciones mixtas hace necesario desarrollar y demostrar la eficiencia de construcciones que activen el RNAi, no solo con secuencias homólogas, sino también con secuencias heterólogas. En el presente estudio se analizó la eficacia de la resistencia inducida a PepGMV en plantas de Nicotiana benthamiana por agroinfiltración de dos construcciones: la primera expresa una dsRNA de la secuencia AV1-IR-AC1 de PepGMV, denominada homóloga (CIRP); la segunda expresa la secuencia AV1-IR-AC1, derivada de ToChLPV y nombrada heteróloga (CIRT). Las plantas protegidas con CIRT presentaron un 45% de eficacia para disminuir la severidad de los síntomas, mientras que las plantas protegidas con CIRP, un 80%. Por otra parte, las plantas protegidas con CIRT mostraron una reducción en la incidencia de 42.86%, mientras que la eficacia en la incidencia en las plantas protegidas con CIRP fue de 57.2%. En cuanto a la eficacia para la carga viral, ésta fue del 95.6% en las plantas protegidas con CIRT y 99.56% en las plantas protegidas con CIRP. The gene silencing mechanism or RNA interference (RNAi) has been used successfully to control diseases caused by these begomoviruses. Strategies of genetic engineering to induce resistance to begomoviruses have used constructions with sequences homologous to the challenged virus. However, the presence of mixed infections, need the development of type RNAi construct to activate the efficiency not only with homologous sequences but also with heterologous sequences. This study analyzed the efficacy of induced resistance to PepGMV in Nicotiana benthamiana plants by agroinfiltration of two constructs: a homologous construct (CIRP) that expresses the dsRNA type hairpin of the sequence AC1-IR-AV1 of PepGMV and a heterologous construct (CIRT) with the sequence AC1-IR-AV1 derived of ToChLPV. Plants rotected with CIRT showed an efficacy of 45% at decreasing the severity of symptoms, whereas plants protected with CIRP showed an efficacy of 80%. Plants protected with CIRT showed a reduction of 42.86%in the incidence, while the incidence in plants protected with CIRP was 57.2%. The efficacy for reducing viral load was 95.6 % in plants protected with CIRT and 99.56 % in plants protected with CIRP.
